Elms are threatened by Dutch elm disease, and conservation methods needed to protect their genetic diversity. Cryopreservation of dormant buds allows large numbers genotypes be conserved with small space requirements minimal upkeep. through slow controlled cooling was tested for both species native Finland, Ulmus glabra laevis. Regeneration the thawed micropropagation studied on different basal media using growth regulators. Multiple surface sterilisation were tried out bud explants. The multiplication U. investigated Driver Kuniyuki walnut medium either 0.5 mg/L meta-topolin or 6-benzylaminopurine. Rooting short indole-6-butyric acid induction in liquid direct transplantation shoots peat ex vitro after tested. For initiation, Murashige Skoog 0.02 gibberellic 4 + 7 6-benzylaminopurine found best promote shoot formation. Surface remains most challenging step. No significant differences between production rooting success. transplanting achieved species, but further development is required before application a larger scale. With improvements success especially glabra, method can applied resources laevis knowledge regeneration used design cryoconservation plan optimise sampling.
